Design Notes
1.Does overlong sequence affect bacterial metabolic activity? 2.The upstream of the lysis gene needs to design the RBS region. 3.When linking gene fragments, homologous arms need to be designed on primers.

The light-controlled genes such as J23100 promoter, yf1 protein and FixJ protein come from the plasmid dusk-egfp-puc57 stored at E. coli from 2018-zjut, while the repression system of lactose operon come from pET28b. The most important gene, lysis,also comes from plasmid dusk-egfp-puc57.
